Our research interest is to study the structures of large protein complexes, molecular machines and membrane proteins, and to elucidate the molecular mechanisms underlying their cellular functions. Single-particle cryo-electron microscopy (cryo-EM) is the major technique in our research work because of its power to determine the three-dimensional structures of large macromolecular assemblies to high resolution, even atomic-resolution, and to analysis their conformational dynamics in near-native environment. Using this technique in combination with biochemical, biophysical and cellular methods, our lab currently focuses on: (1) structures, functions and mechanisms of gigantic protein complexes, macromolecular machines and membrane proteins, (2) molecular mechanisms of membrane trafficking, lipid-protein interactions and membrane-related human diseases.
